Νovel methylation biomarkers in liquid biopsy and classifying biosignatures for the clinical management of breast cancer
Background: Breast Cancer (BrCa) remains a devastating disease presenting emerging needs for effective management. Recently, epigenetic biomarkers are assessed in liquid biopsy for diagnostic and prognostic applications. This study applies a 3-step data-driven biomarker discovery pipeline to identify robust methylation biomarkers and generate high-performance biosignatures specific for clinically significant BrCa end-points, followed by laboratory validation in patient cell-free DNA (cfDNA) … Continua a leggere
Liquid biopsy in Oncology: Results of a Delphi consensus study endorsed by the AIOM-SIAPEC/IAP-SIBioC-SIF Italian scientific societies
Liquid biopsy (LB) offers a minimally invasive alternative to tissue biopsy by detecting tumor-derived analytes in biological fluids. Nonetheless, its adoption is limited by variability in methodologies. Therefore, this study used a modified RAND/UCLA approach involving 23 experts of the field, providing two questionnaires that assessed agreement on 22 items. Consensus was reached for all the pre- and post-analytical phase items, agreeing on the pivotal role of plasma cfDNA. Conversely, opinions varied regarding other biomarkers and biological samples … Continua a leggere
Sacituzumab Govitecan plus Pembrolizumab for Advanced Triple-Negative Breast Cancer
Background: Triple-negative breast cancer is an aggressive breast cancer subtype, and there remains an unmet need to improve outcomes in patients with previously untreated, programmed death ligand 1 (PD-L1)-positive, locally advanced unresectable or metastatic triple-negative breast cancer. Methods: In this phase 3, open-label, international trial, we randomly assigned patients in a 1:1 ratio to receive sacituzumab govitecan plus pembrolizumab or chemotherapy plus pembrolizumab … Continua a leggere
Neoadjuvant Taxane Plus Trastuzumab and Pertuzumab With or Without Carboplatin in Human Epidermal Growth Factor Receptor 2-Positive Breast Cancer: The Randomized Noninferiority Phase III neoCARHP Trial
Purpose: The neoCARHP aimed to investigate the efficacy and safety of investigator-selected taxane (docetaxel, paclitaxel, or nab-paclitaxel) plus trastuzumab and pertuzumab, with carboplatin (TCbHP) or without carboplatin (THP), in stage II and III human epidermal growth factor receptor 2 (HER2)-positive breast cancer … Continua a leggere
Extended Endocrine Therapy Following 5 Years of Adjuvant Luteinizing Hormone-Releasing Hormone Agonist in Premenopausal Patients With Node-Positive, Hormone Receptor-Positive Breast Cancer: A Cohort Study
Purpose: To evaluate the clinical benefit of extended endocrine therapy (eET) after 5 years of adjuvant treatment with luteinizing hormone-releasing hormone agonists (LHRHa) in premenopausal women with node-positive, hormone receptor-positive early breast cancer (eBC). Methods: We conducted a cohort study analysis on two prospectively collected data sets (the Young Women’s Breast Cancer Study and IEO Breast Cancer Cohort) … Continua a leggere
